I've been reading through the classics, and I would say all three that I just finished:

1. Akira

2. A Drifting Life

3. A Drunken Dream and Other Stories

In the way of anime, which I haven't watched much of, Akira, Code Geass, and Mushi-shi are very high up on the list. I've also heard good things about Stargazing Dog and a new one which I've got on hold at the local library, A Zoo in Winter. If you liked Bakuman, this is a better version (apparently).

I have finished watching Darker Than Black seasons 1 and 2, and the Gaiden OVAs.
Damn but the story is depressing. I knew this director liked his story arcs tragic (Wolf's Rain, anyone?), why did I ever delude myself into thinking that I wanted to know what eventually happened to the characters I fell in love with?
Some blogger remarked that season 1 is like watching Ocean's 11, while season 2 is like watching The Professional. Well, I 100% agree, while adding the words "relentlessly tragic" to both descriptions. I feel so bad for all the major characters. :(

That's easy - Space Battleship Yamato. Never, EVER ask me about Space Battleship Yamato. :D My wife and I even just watched the live action version this past December (See my full review of it here) and it was awesome. I was very happy to be in there when Japanese fantasy was being first introduced to the US, and for me, "anime" is still Yamato, Speed Racer, and Battle of the Planets... with a lot of Ultraman mixed in.

Love Hina would be an excellent one for this. A romantic comedy that has plenty of slap-stick wacky moments the whole way through.

There's also Bludgeoning Angel Dokuro-Chan, which is...not sure I can explain it, really...

For something just flat-out off-the-wall wacky, FLCL non-stop.

Azumanga Daioh can't get enough recommendations from me, especially for people wanting something to laugh at without worrying too much about some overarching plot. :)

Full Metal Panic: Fumoffu is another laugh-a-minute series, and of course another of my personal favorites.

GTO/Great Teach Onizuka is hilarious if you don't mind a lot of action and general awesomeness and epicness mixed in with the comedy.

I My Me! Strawberry Eggs is another one I can recommend fondly. A man wanting to be a teacher? Check. School hiring? Check. School only hires female teachers? Che-whoops...what to do? Hilarity and heartwarming ensues.

Those Who Hunt Elves, a series about a group of humans out to return home by finding magic symbols scattered on elven bodies. In other words: A series created as an excuse to strip elves.

Slayers is a good series for mixing high fantasy with the comedy. Lots of good action and plenty of laughs.

Project A-ko is a good series of movies, and was my personal introduction to anime way back in 1991 or so, when the Sci-Fi Channel had their first...I think they called it a 'festival of animation.' A classic even to this day.

There are many more and plenty of us should have a lot to offer. I'm going to call it here for the moment though just to keep from going on for too long. :)

I figured I'd update with one I just found on Crackle on Xbox Live.

Ore no Imouto ga Konna ni Kawaii Wake ga Nai/My Sister Can't Be This Cute.

This is the story of a guy who just wants to live a plain, average, ordinary, uneventful life.

This is the story of a guy who has an over-achieving little sister whose high marks mean people expect from form him, thus making such a life difficult.

This is the story of a guy who finds out his little sister has a secret she can't tell anyone else.

This is the story that is the first in a long time provide me three spit-takes within two episodes. Hell, I'm still watching the second episode. So far...this one is looking to go on my highly-recommended list of comedies.

Luna and Frosty are totally overloading your brain with recommendations.
I'll give you 2:

(1) FLCL -- Agreed. This is a classic. It's wacky/ off-the-wall. It's (somewhat) deep. Aaaand it's short, so low investment.
(2) The Melancholy of Haruhi Suzumiya -- I'm surprised the other 2 haven't mentioned this one. It's a classic. It's wacky/ off-the-wall. It's got an intelligent male protag (quite uncommon). It's great.

I would also recommend checking out some of the Studio Ghibli films if you are starting out. They introduce you a bit to the style of an anime and some of the quirks you often see, but are fairly accessible and do not require the same devotion of time as an actual series. The ones I like the best are Princess Mononoke, Howl's Moving Castle and Naussica Valley of the Wind (Naussica is from the 80s and is a bit dated in some of the choices made).

^^^ This. Seriously. The Miyazaki movies are still probably the best anime I have seen. The really early one, Lupin IIRC, is a hoot, and my own faves are Kiki's Delivery Service, Porco Rosso, and Spirited Away. The Ghibli movies are nearly impossible not to like, and are a great way to introduce non-anime fans to anime.

Trust me, I debated whether to recommend it or not. Ultimately I didn't because it's not a straight-forward comedy and requires a lot more attention paid than a straight-up comedy, something one doesn't really have to worry about with the rest of the choices I put up. :)

The rest of my recommendations are the sort one can pop in, sit back, and relax with. Haruhi really demands some level of attention paid. They put in excessive detail to the backstory, the background (both characters, and actual background in the scenes), and even all the theories and explanations for things that get thrown around.

It is funny, but it has a much more serious side, and as much as I love the series (It is number 2 on my all-time favorites list) I can't recommend it to someone just getting into anime. I would save this one for after a person finds their niche. Then, whatever that niche may be, give it a watch. It does fit into most of them.

Otherwise, yeah, it would've been tops on my list up there. I can't really speak highly enough about the series itself, and find myself praying for a third season to the anime. The movie really leaves things ready for it.

Definitely Nichijou or Azmungah Daioh. Azmungah Daioh is more of a starting point. Watch Nichijou as an anime.
